We put the Mayo Clinic Proceedings video first because it is short and because its fact-check teaches the most important habit for this molecule: check the label before believing a use is established. Tirzepatide is approved as Mounjaro for type 2 diabetes and as Zepbound for chronic weight management and obstructive sleep apnoea; the study in that video is off-label and retrospective. The Endocrine Society journal club follows for viewers who want the pharmacology of dual agonism. The CMHC conversation with Dr. Juan Frias is the practical core: dose steps, switching, the contraceptive caution, cardiovascular data as of 2023. Numbers to carry with you
SURMOUNT-1, 72 weeks, adults without diabetes: mean weight change of 15.0, 19.5 and 20.9 percent on 5, 10 and 15 mg against 3.1 percent on placebo (PubMed 35658024). Label escalation: 2.5 mg for four weeks, then 2.5 mg steps at least four weeks apart to a maximum of 15 mg. Most common adverse reactions in the Zepbound label: nausea 25 to 29 percent, diarrhoea 19 to 23 percent, constipation 11 to 17 percent, vomiting 8 to 13 percent, against single-digit to low-teen placebo rates.
